About Our Technology

The GeoMx Digital Spatial Profiler developed by NanoString is a cutting-edge technology that allows for thorough analysis of RNA and protein in tissue samples. This is achieved by staining the tissue slides with a variety of biological probes, each containing a specific DNA barcode that can be cleaved with UV light. Additionally, fluorescent markers are included to aid visual or computerized interpretation of tissue morphology. By selecting regions of interest (ROIs) based on morphology markers and illuminating them with UV light, the cleaved barcodes can be counted using the NGS platform.

How Does GeoMx Work?

The GeoMx platform allows for the profiling of gene expression in-situ, meaning that gene expression can be analyzed directly within intact tissue sections. This is accomplished through the integration of traditional histology methods with high-throughput molecular profiling. By utilizing a spatially resolved assay, the platform captures and measures RNA transcripts from individual cells in a tissue section.

GeoMx allows researchers to obtain insights about the spatial distribution of gene expression patterns, cellular diversity, and interactions within tissues. This technology is a useful resource for studying intricate biological mechanisms, including growth, disease advancement, and reaction to therapy.

Researchers can gain a more comprehensive understanding of cellular interactions, tissue organization and the underlying mechanisms driving disease or normal physiological processes by combining spatial transcriptomics with traditional histology. The sample is first hybridised with probes that have unique DNA indexing oligonucleotides via a UV photocleavable linker 1.

Researchers select Regions of Interest (ROI) on the GeoMX DSP based on morphological staining (Immunofluorescence or RNAscope) and the DSP barcodes are UV cleaved and collected 2-5.

The sequencing library is constructed with DSP barcodes and ROI specific indexes 6.

The sequencing results are imported back into the GeoMX software for integration with the slide images.
